H-R3 Plus Radiation Responses in Advanced Head and Neck Cancer

Researchers from India have reported that the addition of nimotuzumab to radiation therapy or radiation therapy plus chemotherapy increases long-term loco-regional control and survival in patients with advanced head and neck cancer.

The details of this Phase IIB study were presented at the 2009 meeting of the American Society for Therapeutic Radiation and Oncology (ASTRO).

Nimotuzumab is a humanized anti-epidermal growth factor receptor antibody (EGFR), which was developed in Cuba and is apparently approved in several countries for the treatment of head and neck cancer. There has been suggestive data that nimotuzumab has significant activity in patients with relapsed or refractory gliomas.  A study from Cuba reported a response rate of 38% in patients with newly diagnosed gliomas.

The current study included 92 patients with advanced head and neck cancer treated between 2004 and 2005. These patients were randomly allocated to one of four treatments:

  • Radiotherapy alone
  • Radiotherapy with nimotuzumab
  • Chemoradiotherapy
  • Chemoradiotherapy with nimotuzumab


These authors concluded that the addition of nimotuzumab to radiation therapy or to chemoratiotherapy improved outcomes of patients with advanced head and neck cancer.
